AffiASSAY® Catalase Activity Colorimetric Assay Kit
AffiASSAY® Catalase Activity Colorimetric Assay Kit is designed to provide precise and reliable measurements of catalase enzyme activity in biological samples. This assay kit offers a colorimetric detection method, making it a valuable tool for researchers in the fields of biochemistry, cell biology, and oxidative stress research.
Key Features:
- High Sensitivity: Our assay kit is optimized to detect catalase activity, allowing for accurate quantification in a wide range of samples.
- Colorimetric Detection: Utilizing a highly sensitive colorimetric assay, our kit ensures robust and reproducible results, minimizing the margin for error.
- Wide Application: The AffiASSAY® Catalase Activity Colorimetric Assay Kit can be used with various sample types, including cell lysates, tissue homogenates, blood, serum, plasma, and culture supernatants.
- Quick and Easy: With a simple and streamlined protocol, you can obtain results in just a few easy steps, saving you valuable time in the lab.
- Versatile: Whether you're studying oxidative stress, assessing antioxidant enzyme activity, or investigating the effects of drug compounds, our kit provides the versatility you need.
Assay Principle:
This assay kit measures catalase activity based on the decomposition of hydrogen peroxide (H2O2) by catalase enzyme. The reaction produces water and oxygen, and the decrease in H2O2 concentration is monitored by the decrease in absorbance at a specific wavelength, allowing for the quantification of catalase activity.
Applications:
- Oxidative Stress Research: Assess catalase activity as part of oxidative stress studies to understand the balance between reactive oxygen species (ROS) production and antioxidant defense mechanisms.
- Cell Biology: Measure catalase activity in cell lysates or culture supernatants to evaluate cellular antioxidant capacity and response to oxidative insults.
- Biomedical Research: Investigate the role of catalase in various physiological processes, including inflammation, aging, and disease states such as cancer and neurodegenerative disorders.

AffiASSAY® Catalase Activity Colorimetric Assay Kit Protocol:
[Please note that this protocol is a general guideline, and you should always refer to the specific instructions provided in the kit manual for accurate and detailed information.]
Materials Needed:
- AffiASSAY® Catalase Activity Colorimetric Assay Kit
- Biological samples containing catalase
- 96-well microplate
- Microplate reader capable of measuring absorbance at specific wavelengths
- Pipettes and tips
- Microcentrifuge tubes
Protocol:
Standard Curve Preparation:
- Reconstitute the Catalase Standard provided in the kit according to the kit's instructions.
- Create a series of standard dilutions using the reconstituted Catalase Standard to generate a standard curve. Typical concentrations may range from 0 to 1000 U/mL.
Assay Plate Setup:
- Label the microplate wells according to your experimental design, including wells for your samples, standards, and blanks.
- Prepare blank wells containing assay buffer only.
Sample and Standard Addition:
- Add 50-100 µL of your prepared samples and Catalase Standards to the appropriate wells on the microplate.
- Ensure that the final reaction volume in each well is consistent.
Reaction Initiation:
- Add 50 µL of Catalase Substrate (Hydrogen Peroxide) to each well to initiate the reaction.
Reaction Termination:
- Add 50 µL of Potassium Hydroxide (KOH) to each well after a specified incubation period to stop the reaction.
Measurement:
- Measure the absorbance of each well at a specific wavelength (provided by the kit) using a microplate reader.
Data Analysis:
- Generate a standard curve using the absorbance values obtained from the Catalase Standards.
- Calculate the catalase activity in your samples based on their respective absorbance readings and the standard curve.
Results Interpretation:
- Interpret and analyze your results according to your research objectives.
- Report catalase activity as units per milliliter (U/mL) or units per milligram of protein (U/mg protein) if protein normalization is performed.
Data Presentation:
- Prepare your data for presentation or publication, and include the appropriate statistical analysis if required.
Cleanup:
- Dispose of all waste materials, including used assay components, according to your laboratory's safety protocols.
- Always refer to the specific instructions provided with the AffiASSAY® Catalase Activity Colorimetric Assay Kit for any kit-specific details or variations in the protocol.
- Additionally, ensure that you adhere to the best practices and safety guidelines in your laboratory while conducting the assay.
