Product Insert 

Test Principal:

  • The AffiVET® CPV Antibody Rapid Test Kit employs a chromatographic immunoassay for the semiquantitative detection of IgG antibodies to parvovirus in canine serum, plasma, or whole blood.
  • This kit features a T-line and a C-line on the device's surface, initially invisible before sample application.
  • The appearance of the C-line serves as a reference, confirming the test's proper functionality and should consistently appear with each test. In the presence of IgG antibodies specific to canine parvovirus in the sample, a purple T-line will appear in the result window.
  • The assay utilizes highly selective antigens as both capture and detector components, ensuring precise detection of IgG antibodies to canine parvovirus in samples.

Storage & Stability:

  • Store the test kit at 2~30°C. DO NOT FREEZE.
  • Do not store the test kit in the direct sunlight.
  • The test kit is stable within the expiration date marked on the package label.

Sample collection & Preparation:

1. Use canine whole blood, serum, or plasma with this test. For whole blood: Collect the blood into the provided anticoagulant tube (maximum volume 1.5ml). If not used immediately or within 24 hours, refrigerate at 2~8°C.
For serum: Collect blood into a collection tube without anticoagulants, allow it to settle for 30 minutes for coagulation, then centrifuge to obtain serum.
For plasma: Collect blood into a collection tube with anticoagulants, then centrifuge to obtain plasma.
2.  If serum or plasma samples are not tested immediately, they should be refrigerated at 2~8°C. For longer storage, freezing is recommended. Frozen samples should be brought to room temperature (15~30°C) prior to use.
3.  Clarify samples containing precipitate before testing to avoid inconsistent results.
4.  Avoid using hemolytic, lipemic, icteric, or bacterially contaminated samples as they may lead to erroneous results.

 

Test Procedure:

1.  Allow all kit components and samples to reach room temperature before conducting the test.
2.  Whole blood: Place a drop of whole blood on the specimen sheet, then use an inverted cup to collect 5μl of the sample. The whole blood will be automatically drawn into the inverted cup due to surface tension.
Serum/Plasma: Collect 5μl of the serum or plasma sample using a micropipette (not provided).
3.  Add the sample into the assay diluent tube.
4.  Mix the inverted cup until the sample has completely dissolved into the assay diluent.
5.  Remove the test kit from the foil pouch before using it.
6.  Using the disposable dropper provided, transfer the samples into the tube
7.  Add four (4) drops of the sample into the sample hole on the test device, dropping each drop vertically one by one.
8.  Start the timer and observe as the sample flows across the result window. If the sample does not appear after 1 minute, add one more drop of the prepared sample to the sample hole
9.  Interpret the test results precisely at 10 minutes after starting the test. Do not interpret the results after 20 minutes as they may not be accurate or reliable.

    Result Interpretation:

    Negative result

    Presence of C-Line
    Absence of T-Line

    Positive result:

    Presence of C-Line
    Presence of T-Line

    The color development of the T-line is weaker than that of the C-line, indicating a low titer below 1:40 as per the HI titer, falling within the color scale range of 1~2.

    Antibody titer is low against CPV.

    The T-line shows medium titer (1:80 as HI titer) with color development equal to that of the C-line, within the color scale of 3.

     

     

    Antibody titer is medium against CPV.

    This is indicative of a good immune status.

     

    The T-line indicates a high titer (above 1:160 as per the HI titer) with significantly stronger color development compared to the C-line, within the color scale of 4~6.

     

    Antibody titer is high against CPV.
    This is indicative of a good immune status.

    Invalid Result:

    Absence of C-Line
    Presence of T-Line

    Absence of C-Line

    Cautions:

    • Use this test kit exclusively for canine samples; do not use it for other animals.
    • The test device is sensitive to humidity and heat, so perform the test immediately after removing it from the foil pouch.
    • Do not reuse any test components to ensure accurate results.
    • Apply the sample using a disposable dropper in a vertical manner.
    • Avoid touching the membrane in the result window of the test device.
    • Do not use the test kit beyond its expiration date indicated on the package label.
    • Do not use the test kit if the pouch is damaged or if the seal is broken.
    • Refrain from mixing components from different lot numbers, as each kit undergoes quality control testing as a standard batch unit.
    • Treat all samples as potentially infectious and wear protective gloves during handling. Thoroughly wash hands afterward.
    • Dispose of all samples, reaction kits, and potentially contaminated materials safely in accordance with national and local regulations.
