AffiAB® Anti-dsRNA Monoclonal Antibody [J2] (Mouse IgG2a, κ)

The AffiAB® Anti-dsRNA Monoclonal Antibody J2 is widely regarded as the gold standard for the detection of double-stranded RNA (dsRNA). It is validated for use in a variety of applications including cultured cells and fixed paraffin-embedded tissue samples.

Note: For dsRNA immunoblotting, nucleic acid separation must be performed using polyacrylamide gel electrophoresis. Sensitivity is significantly reduced when transferred from agarose gels.

Specificity:

• Recognizes double-stranded RNA (dsRNA) with helix lengths ≥40 base pairs.
• Recognition is sequence- and nucleotide-independent.
• Detects a broad range of natural dsRNAs (40–50 species), as well as synthetic analogs such as poly(I):poly(C) and poly(A):poly(U).
• Affinity for poly(I):poly(C) is approximately 10-fold lower compared to other dsRNA targets.

Formulation & Reconstitution:

• Form: Lyophilized from 1 mg/mL PBS solution
• Solubility:
  To prepare a 1 µg/µL antibody solution:

1. Add 200 µL of sterile, DNAse/RNAse-free distilled water to vial AFG-JBS-0002 200 µg, or
2. Add 500 µL to vial AFG-JBS-0002 500 µg. 

Important: Due to the lyophilization process, minor amounts of denatured protein aggregates may form upon reconstitution. These may affect certain applications (e.g., immunohistochemistry). We recommend centrifuging the reconstituted antibody and using the supernatant only.

• Storage Conditions:

1. Store at -20°C to -80°C after reconstitution for long-term stability.
2. Avoid freeze-thaw cycles; aliquot upon reconstitution.
3. With the addition of 10 mM sodium azide, the undiluted antibody (1 µg/µL) may be stored at 4°C for short-term use.

• Shelf Life: 12 months from the date of delivery.

• Purity & Identity: Verified via reducing and non-reducing SDS-PAGE
• Functional Activity: Evaluated by AN-ELISA, compared to a J2 reference standard.
