AffiVET® Poultry Newcastle Disease NDV Antigen Rapid Test Kit
Rapid chromatographic immunoassay for the qualitative detection of Newcastle Disease Virus antigen in chicken samples.
Product Name
AffiVET® Poultry Newcastle Disease NDV Antigen Rapid Test Kit
Catalog Number
AFG-VR-76
Package Size
10 Tests/Kit
Intended Use
The AffiGEN AffiVET Rapid NDV Ag Test Kit is a rapid chromatographic immunoassay for the qualitative detection of Newcastle Disease Virus antigen, also known as NDV Ag, in chicken samples. The test is intended as a rapid aid in the detection of NDV antigen.
| Sample Type | Use |
|---|---|
| Oropharyngeal swab | Recommended |
| Tracheal swab | Suitable |
| Spleen sample / homogenate | Suitable |
| Kidney sample / homogenate | Suitable |
Principle of the Test
The test uses NDV-specific antibodies as capture and detector antibodies. When NDV antigen is present in the sample, a visible test line, marked T, appears in the result window. The control line, marked C, confirms that the test has run correctly.

Materials Required but Not Provided
| Material | Purpose |
|---|---|
| Timer | Reading the result at the correct time |
| PBS | For spleen or kidney homogenate preparation, if required |
| Protective gloves | Safe sample handling |
Storage and Stability
- Store the kit at 2–30°C.

Sample Collection and Preparation
Swab Samples
Collect an oropharyngeal or tracheal swab from the chicken. Use the swab immediately when possible.
Spleen or Kidney Samples
Prepare a 20–50% homogenate using PBS.
| Homogenate Preparation | Example |
|---|---|
| 50% homogenate | 2 g tissue + 2 mL PBS |
| 20% homogenate | 1 g tissue + 4 mL PBS |
Sample Storage
| Storage Condition | Maximum Duration |
|---|---|
| 2–8°C | Up to 24 hours |
| ≤ -20°C | Longer storage |
Allow frozen samples to reach room temperature before testing.
Test Procedure
Interpretation of Results
| Result | Appearance | Interpretation |
|---|---|---|
| Negative | C line only | NDV antigen not detected |
| Positive | C line + T line | NDV antigen detected |
| Invalid | No C line | Test invalid; repeat with a new device |
Performance Characteristics
| Parameter | Reported Value |
|---|---|
| Sensitivity vs RT-PCR | 94.7% |
| Specificity vs RT-PCR | 96.4% |
| Detection limit | Approx. 0.25 HAU |
Comparison with RT-PCR
| RT-PCR Positive | RT-PCR Negative | |
|---|---|---|
| AffiVET Rapid NDV Ag Positive | 18 | 3 |
| AffiVET Rapid NDV Ag Negative | 1 | 81 |
The product sheet also reports no cross-reaction with other avian viruses and states that the test detects all NDV.
Limitations
- A negative result does not fully exclude NDV infection, especially when antigen levels are below the detection limit or when sample quality is poor.
- Final diagnosis should not be based only on a single rapid test result. Clinical signs, flock history, epidemiological information, and additional laboratory testing should be considered when needed.
- False positive or false negative results may occur rarely.
Precautions
- Use only with chicken samples.
- Do not use expired kits.
- Do not use a test device if the foil pouch is damaged.
- Do not reuse test devices, swabs, droppers, or diluent tubes.
- Do not mix components from different lot numbers.
- Handle all samples as potentially infectious.
- Dispose of used materials according to local biosafety regulations.
