Test Principal:

The AffiVET® Rapid AIV Ag Test Kit uses sandwich chromatographic immunoassay technology to qualitatively detect avian influenza type A NP antigen in chicken or duck (excluding wild animal) cloaca discharge and tracheal secretions.

The device features a test (T) line and a control (C) line, where the control line serves as a reference ensuring the test is functioning correctly. The appearance of the control line confirms proper test execution.

Highly selective antibodies specific to avian influenza type A NP antigen are used in both capture and detection roles within the assay.

Storage & Stability:

  • Store the test kit at 2~30℃. DO NOT FREEZE.
  • Protect the kit from light and store it in dry environment.
  • The test kit is stable within the expiration date marked on the package label.

Sample collection & Preparation:

  1. Use samples from avian tracheal secretions, cloacal discharge to ensure accurate detection of AIV  antigen.
  2. If the sample cannot be tested immediately, refrigerate it at 2-8°C for up to 24 hours. For longer storage, freeze the sample at -20°C or below.  Frozen samples should be brought to room temperature (15~30°C) prior to use.

Test Procedure:

  1. Bring all kit components and samples to room temperature before use to ensure optimal performance.
  2. Collect samples from poultry laryngeal tracheal secretions and cloacal discharges.
  3. Insert the swab into parts with most secretions and gently swipe the swap to make it fully absorb the secretions.
  4. Insert the prepared swab into the sample buffer tube provided in the kit.
  5. Drain the swab after stir by squeeze the liquid out against the wall of the tube.
  6. Remove the test device from the foil pouch and place it on a flat, dry surface.
  7. Using the disposable pipette provided, extract the supernatant sample from the tube.
  8. Apply 3 to 4 drops of the sample into the sample hole on the test device, adding each drop one at a time.
  9. Start the timer and observe as the sample flows across the result window. If the sample does not appear after 1 minute, add one more drop of the prepared sample to the sample hole.
  10. Interpret test results at 15 minutes. Do not read the results after 20 minutes.

Result Interpretation:

Negative result

Presence of C-Line
Absence of T-Line

This means that there is no AIV antigens in the sample or the level is lower than the test detection limit.

Positive result:

Presence of C-Line
Presence of T-Line

This confirms the presence of the AIV antigens in the sample.

Invalid Result:

Absence of C-Line
Presence of T-Line

Or

Absence of C-Line
Absence of T-Line

This means that the procedure was not applied correctly or that the test is invalid.

Test Precautions:

  • The AffiVET® Rapid AIV Ag Test Kit is intended for use with avian samples only, excluding wild animals. Do not use the test for other animals.
  • The test device is sensitive to both humidity and heat. Perform the test immediately after removing the test device from the foil pouch.
  • Do not reuse any test components.
  • Apply the assay diluent vertically.
  • Avoid touching the membrane within the result window of the test device.
    Do not use the test kit beyond the stated expiration date marked on the package label.
  • Do not use the test kit if the pouch is damaged or if the seal is broken.
  • Do not mix components from different lot numbers. The components in this kit have been quality control tested as a standard batch unit.
  • Handle all samples as potentially infectious. Wear protective gloves when handling samples and wash hands thoroughly afterward.
  • Decontaminate and dispose of all samples, used kits, and potentially contaminated materials safely in accordance with national and local regulations.

Test Limitations:

  • AffiVET® Rapid AIV Ag Test kit is highly accurate in detecting  avian influenza virus type A antigen. However, occasional false results may occur.
  • Positive result shall be further confirmed by PCR and/or other official methodologies. Diagnosis of Influenza shall be determined by the vet, considering both clinical symptoms and laboratory tests.
  • The Color of Line T is related to influenza antigen contained in the sample. If the antigen concentration is at very high level, Line T may appear and then be lighter, in this case, please future dilute the sample solution with DD water and run the test again to see a stable Line T.
