Description
The AffiAB® Monoclonal dsRNA Antibody Panel includes the J2, J5, and K1 antibodies, each specifically developed for the high-affinity recognition of double-stranded RNA (dsRNA) structures longer than 40 base pairs, independent of sequence or nucleotide composition. These antibodies have been widely used to detect dsRNA intermediates of RNA viruses and synthetic analogs such as poly(I:C).

• J2: The most commonly used anti-dsRNA antibody for general applications.
• J5: Shares similar specificity and affinity to J2 but is of a different isotype (IgG2b), ideal for multiplex detection.
• K1: Preferred for detecting poly(I:C) due to its higher affinity for this synthetic analog compared to J2 or J5.

These antibodies can be used in a broad range of applications, including:

Note: For dsRNA immunoblotting, polyacrylamide gel electrophoresis (PAGE) is required. Detection sensitivity is substantially reduced after blotting from agarose gels.

Storage Conditions

• Upon reconstitution: store at –20 °C to –80 °C for long-term storage.
• Avoid repeated freeze/thaw cycles; store in aliquots.
• After reconstitution and addition of 10 mM sodium azide, the antibody (1 μg/μl) can be stored at 4 °C for short-term use.

Solubility and Reconstitution
Reconstitute each vial in:

• 100 μL sterile, DNase/RNase-free distilled water to prepare a 1 μg/μL solution in PBS.

Due to lyophilization, minor aggregation may occur. For optimal results:

• Centrifuge the reconstituted antibody in a microcentrifuge.
• Use the supernatant only to avoid interference in sensitive applications (e.g., IHC).

12 months from the date of delivery (lyophilised)

Specificity
All three antibodies (J2, J5, K1) detect dsRNA helices ≥ 40 bp. They do not cross-react with ssRNA, ssDNA, or dsDNA.

Quality Control

• Purity & Identity: Verified by reducing and non-reducing SDS-PAGE
• Activity: Verified by Anti-Nucleic acid ELISA (relative to reference K1)
