AffiASSAY® Glucose Colorimetric & Fluorometric Assay Kit

The AffiASSAY® Glucose Colorimetric & Fluorometric Assay Kit offers a reliable method for the quantification of glucose in various biological samples. This versatile assay kit provides flexibility with options for both colorimetric and fluorometric detection, making it suitable for a wide range of research applications in metabolism, diabetes, and glucose homeostasis studies.

Key Features:

  • Dual Detection: Our assay kit enables the measurement of glucose using both colorimetric and fluorometric methods, offering versatility based on experimental needs.
  • High Sensitivity: The kit can detect low levels of glucose, ensuring accurate quantification in biological samples.
  • Wide Application: The AffiASSAY® Glucose Assay Kit is compatible with various sample types, including serum, plasma, urine, cell lysates, and culture media.
  • Quick and Easy: With a streamlined protocol, you can obtain results rapidly, saving time and resources in the laboratory.
  • Cost-Effective: Our kit provides an affordable solution for measuring glucose levels, reducing experimental costs without compromising data quality.

Assay Principle:

The assay kit measures glucose based on the enzymatic oxidation of glucose to generate a colorimetric or fluorometric signal. The signal intensity is directly proportional to the glucose concentration in the sample.

Applications:

  • Metabolism Studies: Investigate glucose metabolism and its role in energy production and storage.
  • Diabetes Research: Study glucose homeostasis, insulin resistance, and diabetes-related complications.
  • Nutritional Studies: Assess the impact of dietary interventions and nutritional supplements on blood glucose levels and metabolic health.

Kit Contents:

  • Glucose Assay Buffer
  • Glucose Enzyme Mix
  • Glucose Probe
  • Microplate (96 wells)
  • Glucose Standard
  • Detailed Protocol

AffiASSAY® Glucose Colorimetric & Fluorometric Assay Kit Protocol:

[Please note that this protocol is a general guideline, and you should always refer to the specific instructions provided in the kit manual for accurate and detailed information.]

Materials Needed:

  • AffiASSAY® Glucose Colorimetric & Fluorometric Assay Kit
  • Biological samples containing glucose
  • Microplate reader capable of measuring absorbance and fluorescence
  • Pipettes and tips
  • Microcentrifuge tubes

Protocol:

Standard Curve Preparation:

  • Prepare a series of standard solutions containing known concentrations of the glucose standard provided in the kit.
  • Dilute the Glucose Probe with Glucose Assay Buffer to prepare the working solution.
  • Add the working solution to each standard solution to initiate the reaction.

Assay Plate Setup:

  • Label the microplate wells according to your experimental design, including wells for your samples, standards, and controls.
  • If using controls, prepare control samples without glucose to serve as background controls.

Sample and Standard Addition:

  • Add 50-100 µL of your prepared samples and standard solutions to the appropriate wells on the microplate.
  • Ensure that the final reaction volume in each well is consistent.

Assay Reaction:

  • Prepare the Glucose Assay Buffer by diluting as instructed in the kit manual.
  • Add the Glucose Enzyme Mix and Glucose Probe to each well to initiate the reaction.
  • Incubate the microplate at the appropriate temperature (as specified in the kit instructions) for a recommended time to allow the enzymatic reaction to proceed.

Colorimetric and Fluorometric Measurement:

  • After the incubation period, measure the absorbance and fluorescence of each well using a microplate reader.
  • For colorimetric detection, measure the absorbance at the appropriate wavelength.
  • For fluorometric detection, measure the fluorescence intensity using appropriate excitation and emission wavelengths.

Data Analysis:

  • Generate standard curves using the absorbance or fluorescence values obtained from the standard solutions.
  • Calculate the glucose concentrations in your samples based on their respective absorbance or fluorescence readings and the standard curves.

Results Interpretation:

  • Interpret and analyze your results according to your research objectives.
  • Observe any variations or trends in glucose levels among your samples.

Data Presentation:

  • Prepare your data for presentation or publication, and include the appropriate statistical analysis if required.

Cleanup:

  • Dispose of all waste materials, including used assay components, according to your laboratory's safety protocols.
  • Always refer to the specific instructions provided with the AffiASSAY® Glucose Colorimetric & Fluorometric Assay Kit for any kit-specific details or variations in the protocol.
  • Additionally, ensure that you adhere to the best practices and safety guidelines in your laboratory while conducting the assay.
