AffiASSAY® D-Lactate Colorimetric Assay Kit

The AffiASSAY® D-Lactate Colorimetric Assay Kit provides a precise and reliable method for measuring D-lactate levels in various biological samples. This assay kit offers a simple and efficient solution for researchers in fields such as clinical diagnostics, microbiology, and nutrition.

Key Features:

  • High Sensitivity: Our assay kit can detect low levels of D-lactate, ensuring accurate quantification in biological samples.
  • Colorimetric Detection: Utilizing a colorimetric assay, our kit offers a simple and easy-to-read readout, suitable for both manual and automated analysis.
  • Wide Application: The AffiASSAY® D-Lactate Assay Kit is compatible with various sample types, including serum, plasma, urine, tissue homogenates, and cell lysates.
  • Quick and Easy: With a streamlined protocol, you can obtain results rapidly, saving time and resources in the laboratory.
  • Cost-Effective: Our kit provides an affordable solution for measuring D-lactate levels, reducing experimental costs without compromising data quality.

Assay Principle:

This assay kit measures D-lactate levels based on the enzymatic conversion of D-lactate to generate a product that reacts with a colorimetric probe. The intensity of the color formed is proportional to the D-lactate concentration in the sample.

Applications:

  • Clinical Diagnostics: Measure D-lactate levels in patient samples for the diagnosis and monitoring of lactate acidosis, bacterial infections, and other metabolic disorders.
  • Microbiology: Assess D-lactate production by bacteria and other microorganisms in culture media or biological samples.
  • Nutrition Studies: Evaluate D-lactate levels in food products and beverages to assess fermentation processes and food quality.

Kit Contents:

  • D-Lactate Assay Buffer
  • D-Lactate Enzyme Mix
  • D-Lactate Probe
  • 96-Well Assay Plate
  • Detailed Protocol

AffiASSAY® D-Lactate Colorimetric Assay Kit Protocol:

[Please note that this protocol is a general guideline, and you should always refer to the specific instructions provided in the kit manual for accurate and detailed information.]

Materials Needed:

  • AffiASSAY® D-Lactate Colorimetric Assay Kit
  • Biological samples containing D-lactate
  • 96-well microplate
  • Microplate reader capable of measuring absorbance
  • Pipettes and tips
  • Microcentrifuge tubes

Protocol:

Standard Curve Preparation:

  • Prepare a series of standard solutions containing known concentrations of D-lactate.
  • Dilute the D-Lactate Probe provided in the kit with the Assay Buffer to prepare the working solution.
  • Add the working solution to each standard solution to initiate the reaction.

Assay Plate Setup:

  • Label the microplate wells according to your experimental design, including wells for your samples, standards, and controls.
  • If using controls, prepare control samples without D-lactate to serve as background controls.

Sample and Standard Addition:

  • Add 50-100 µL of your prepared samples and standard solutions to the appropriate wells on the microplate.
  • Ensure that the final reaction volume in each well is consistent.

Assay Reaction:

  • Add 50 µL of D-Lactate Enzyme Mix to each well to initiate the reaction.
  • Incubate the microplate at the appropriate temperature (as specified in the kit instructions) for a recommended time to allow the enzymatic reactions to occur.

Colorimetric Measurement:

  • After the incubation period, measure the absorbance of each well at the appropriate wavelength using a microplate reader.

Data Analysis:

  • Generate a standard curve using the absorbance values obtained from the standard solutions.
  • Calculate the D-lactate concentrations in your samples based on their respective absorbance readings and the standard curve.

Results Interpretation:

  • Interpret and analyze your results according to your research objectives.
  • Observe any variations or trends in D-lactate levels among your samples.

Data Presentation:

  • Prepare your data for presentation or publication, and include the appropriate statistical analysis if required.

Cleanup:

  • Dispose of all waste materials, including used assay components, according to your laboratory's safety protocols.
  • Always refer to the specific instructions provided with the AffiASSAY® D-Lactate Colorimetric Assay Kit for any kit-specific details or variations in the protocol.
  • Additionally, ensure that you adhere to the best practices and safety guidelines in your laboratory while conducting the assay.
