Test Principal:

The AffiVET® Rapid Bovine Brucella Antibody Test Kit is a chromatographic immunoassay designed for the qualitative detection of antibodies against Brucella abortus in bovine whole blood, plasma, serum, or milk.

This test kit features two distinct lines on its surface: the test (T) line and the control (C) line. Both lines remain invisible until a sample is applied. The control line serves as a reference point, indicating the proper functioning of the test. It should appear every time the test is conducted.

Presence of antibodies against Brucella abortus in the sample is indicated by the appearance of a purple test line in the result window.

Storage & Stability:

  • Store the test kit at 2~30℃. DO NOT FREEZE.
  • Do not store the test kit in the direct sunlight.
  • The test kit is stable within the expiration date marked on the package label.

Sample collection & Preparation:

  • For this test, only bovine whole blood, serum, plasma, or milk should be used.

Whole blood

Collect blood in an anticoagulant tube. If not tested immediately, refrigerate at 2-8°C and use within 24 hours.

Serum

Collect blood in a tube without anticoagulants (e.g., heparin, EDTA, sodium citrate), let it settle for 30 minutes to clot, then centrifuge to obtain serum.

Plasma

Collect blood in a tube with anticoagulants, then centrifuge to obtain plasma.

Milk

Collect milk into a collection tube.

  • If serum, plasma, or milk samples are not tested immediately, refrigerate at 2-8°C. For longer storage, freeze. Before use, thaw frozen samples to room temperature (15-30°C).
  • Samples with precipitates may give unreliable results. Clarify such samples before testing.
  • Avoid using samples that are hemolytic, lipemic, icteric, or contaminated with bacteria, as they may cause incorrect results.

Test Procedure:

  1.  All reagents and samples must be at room temperature (15~30°C) before use.
  2. Remove the test device from the foil pouch, and place it on a flat and dry surface.
  3. Using a disposable capillary tube, dispense 10 µL of sample into sample hole (S) of the test device.
  4. Add 4 drops of assay diluent into the sample hole (S) vertically.
  5. As the test begins to work, you will see purple color move across the result window in the center of the test device. If the migration has not appeared after 1 minute, add one more drop of the assay diluent to the sample well.
  6. Interpret test results at 20 minutes. Do not interpret after 30 minutes.

This confirms the presence of antibodies against Brucella abortus.

This means that the procedure was not applied correctly or that the test is invalid.

Limitations

1) While the AffiVET® Rapid Bovine Brucella Antibody Test Kit is highly accurate in detecting Brucella abortus antibodies, occasional false results may occur. In cases of uncertainty, additional clinical or laboratory tests may be necessary. A conclusive clinical diagnosis should rely on thorough evaluation by a veterinarian, considering all clinical and laboratory findings, rather than solely on the result of a single test.

2) The reading window may display a light pink background coloration, which does not impact the accuracy of the results.

3) AffiGEN Inc. and its distributors cannot be held liable for any consequences resulting from the misuse or misinterpretation of the test results.

Precautions : 

  • Only use this test kit for cows; it's not meant for other animals.

  • Keep the test device away from humidity and heat.

  • Use it right after taking it out of the foil pouch.

  • Don't try to reuse any parts of the test kit.

  • Put the sample and diluent in vertically.

  • Don't touch the window where the result shows up.

  • Don't use the kit after the expiration date on the label.

  • If the pouch looks damaged or opened, don't use the kit.

  • Don't mix parts from different batches. Each kit is tested as a whole.

  • Treat all samples as if they could be infectious.

  • Wear gloves and wash your hands.

  • Dispose of used kits and materials safely according to local rules.
