Assay Principle:

The assay kit measures glycogen content based on the hydrolysis of glycogen to glucose by glycogenase, followed by the enzymatic oxidation of glucose to generate a colorimetric or fluorometric signal. The signal intensity is directly proportional to the glycogen concentration in the sample.

Materials Needed:

  • AffiASSAY® Glycogen Colorimetric & Fluorometric Assay Kit
  • Biological samples containing glycogen
  • Microplate reader capable of measuring absorbance or fluorescence
  • Pipettes and tips
  • Microcentrifuge tubes

Protocol:

Standard Curve Preparation:

  • Prepare a series of standard solutions containing known concentrations of the glucose standard provided in the kit.
  • Dilute the Glycogen Assay Buffer and Glycogenase Enzyme as instructed in the kit manual.
  • Add the standard solutions to each well of the microplate to generate the standard curve.

Assay Plate Setup:

  • Label the microplate wells according to your experimental design, including wells for your samples, standards, and controls.
