Assay Principle:

The assay kit measures glutamate based on the enzymatic oxidation of glutamate by glutamate dehydrogenase (GLDH) to generate a colorimetric signal. The signal intensity is directly proportional to the glutamate concentration in the sample.

Materials Needed:

  • AffiASSAY® Glutamate Colorimetric Assay Kit
  • Biological samples containing glutamate
  • Microplate reader capable of measuring absorbance at 450 nm
  • Pipettes and tips
  • Microcentrifuge tubes

Protocol:

Standard Curve Preparation:

  • Prepare a series of standard solutions containing known concentrations of the glutamate standard provided in the kit.
  • Dilute the Glutamate Probe with Glutamate Assay Buffer to prepare the working solution.
  • Add the working solution to each standard solution to initiate the reaction.

Assay Plate Setup:

  • Label the microplate wells according to your experimental design, including wells for your samples, standards, and controls.
  • If using controls, prepare control samples without glutamate to serve as background controls.

Sample and Standard Addition:
