Test Principal:

AffiVET® Newcastle Disease Virus (NDV) Antigen Test employs a rapid immuno-chromatography technique to detect NDV  antigen in birds' trachea or cloaca (or feces). Upon adding the sample, it migrates along with the colloidal gold-labeled NDV antibody. If NDV antigen is present in the sample, it binds to the antibody on the T line, producing a wine-red color. Conversely, if no NDV antigen is present, no color reaction occurs.
Newcastle Disease, also known as Asian Fowl Plague, is caused by a virus affecting chickens and various bird species. It is an acutely contagious disease characterized mainly by difficulty in breathing, diarrhea, nervous disorders, and mucosal and serosal bleeding. Due to different pathogenic strains, the severity of the disease can vary widely.

Storage & Expiry Date:

  • Store the test kit at 2 to 30°C. DO NOT FREEZE.
  • Allow the aluminum foil bags to return to room temperature before opening them for storage in a cold environment.
  • The test kit remains stable within its 24-month expiration period, with the production date specified on the box.

Sample Collection & Preparation:

  1. Utilize a cloaca (recommended) or trachea for this test.
  2. Test samples immediately after collection.
  3. Collect the sample using a swab from the cloaca (preferred) or trachea.
  4. To young birds, use a dropper to directly collect fresh feces.
  5. Avoid collecting too much feces sample, which can disrupt liquid flow and cause incorrect results.
  6. Insufficient sample may lead to false negatives. Follow instructions for proper collection.

Test Procedure:

  1. Use Swab to collect sample from cloaca (recommended) or trachea, the swab should be inserted into cloaca or trachea, wipe and rotate in inner well repeatedly. To young birds, il can use dropper to collect fresh feces directly.
  2. Immediately insert the swab into the tube with sample buffer. Rotate the swab strongly on the tube well for at least 10 times to mix the solution evenly, ensuring full dissolution of the sample in the sample buffer. Press the swab on the tube well above the liquid level to fully press out the liquid, then discard the swab.
  3. Allow the solution to stand statically for 1-2 minutes to allow larger particles to precipitate.
  4. Remove the test card from the sealed bag and place it on a flat surface. Use a dropper to draw upper-layer bright liquid and slowly drop 5 drops, one by one, onto the sample well marked with "S".
  5. Leave the test card at room temperature for 15-20 minutes, then read the result. The result is considered invalid after 20 minutes.

Result Interpretation:

Negative Result:

  • Presence of C-Line
  • Absence of T-Line
This means that there is no NDV antigens in the sample or the level is lower than the test detection limit.

    Positive Result:

    • Presence of C-Line
    • Presence of T-Line
    This confirms the presence of the NDV antigens in the sample.

      Invalid Result:

      • Absence of C-Line
      • Presence of T-Line

      or

      • Absence of C-Line
      • Absence of T-Line
      This means that the procedure was not applied correctly or that the test is invalid.

        Limitations:

        • This is a qualitative screening test that accurately detects NDV antigen. However, insufficient sample or antigen concentration lower than sensitivity levels may yield negative results. A clear clinical diagnosis should consider all clinical and laboratory phenomena.
        • Compared to similar test cards, this product has a sensitivity of >99% in detecting Newcastle disease virus antigen and does not cross-react with similar symptoms caused by fowl cholera, avian influenza virus, or infectious bronchitis.
        • Positive reactions may occur post-vaccination with attenuated vaccines due to increased virus levels during the immunization process. However, this phenomenon typically disappears 3-10 days after vaccination.The test card can only be used when brought to room temperature.
