  1. Avoid freeze/thaw cycles – store in aliquots
  2. With 10 mM sodium azide added, the undiluted antibody (1 µg/µL) can be stored at 4 °C for short-term use
  • Shelf Life: 12 months from the date of delivery

Formulation & Reconstitution:

  • Form: Lyophilized from a 1 mg/mL solution in PBS
  • Reconstitution Instructions:

To prepare a 1 µg/µL solution in PBS:

  1. Add 200 µL of sterile, DNAse/RNAse-free distilled water to AFG-JBS-0004 200 µg.
  2. Add 500 µL to AFG-JBS-0004 500 µg.

Note: Lyophilization may result in small amounts of denatured protein aggregates, which could interfere with certain applications such as immunohistochemistry by increasing background.
Recommendation: Centrifuge the reconstituted antibody using a microcentrifuge, and use the supernatant only for experimental applications.

Description:
The AffiAB® Anti-dsRNA Monoclonal Antibody K1 is a high-affinity mouse IgG2a antibody specific for double-stranded RNA (dsRNA). It is suitable for detecting dsRNA in cultured cells and fixed paraffin-embedded tissue samples.
K1 demonstrates higher affinity for poly(I:C) than the J2 clone and serves as an excellent alternative in cases where J2 shows cross-reactivity or high background.

Important Note: For dsRNA immunoblotting, use polyacrylamide gel electrophoresis for nucleic acid separation. Blotting from agarose gels significantly reduces detection sensitivity.

Specificity:

  • Recognizes dsRNA helices ≥40 base pairs
  • Sequence- and nucleotide composition-independent binding
  • Detects all tested naturally occurring dsRNAs (40–50 species)
  • Recognizes poly(I):poly(C) and poly(A):poly(U)

Exhibits higher affinity for poly(I):poly(C) compared to other dsRNA species

Binding constants may vary with experimental conditions (Schönborn et al., 1991)
